A microscope is an instrument that can be used to observe small objects, even cells. The image of an object is magnified through at least one lens in the microscope. This lens bends light toward the eye and makes an object appear larger than it actually is.

Light microscopes are essential scientific instruments that allow us to see objects too small for the naked eye. These powerful tools use a series of lenses to magnify tiny specimens, revealing intricate details of cells, microorganisms, and tiny structures that would otherwise remain invisible. In a typical light microscope, light passes through or reflects off a specimen, travels through objective lenses and eyepiece lenses that further enlarge the image. This enables students and scientists to observe specimens at magnifications typically ranging from 40x to 1000x, opening up an entirely new world of observation.

What a Microscope Is Used For in Scientific Discovery

By making the invisible visible, microscopes have allowed scientists to develop cell theory, understand disease-causing microorganisms, and make countless discoveries that have advanced human knowledge. Microscopes are used to magnify and observe tiny objects, organisms, and structures that are too small to be seen with the naked eye, allowing scientists and students to explore and understand the microscopic world.

Using a stereo microscope or compound microscope, students can observe bacteria and protists moving in drops of pond water. Light microscopes allow students to study the intricate patterns on insect wings, or analyze the crystalline structures of minerals.

Specimens Typically Examined with a Light Microscope

Light microscopes are versatile tools used to examine a wide variety of specimens. The following are commonly examined:

  • Prepared slides of plant and animal tissues, revealing cellular structures like cell walls, nuclei, and chloroplasts
  • Microorganisms in pond water samples, including algae, amoebas, and paramecia
  • Blood smears to see different types of blood cells
  • Insect parts and insect wings
  • Textile fibers
  • Thin sections of rocks and minerals
  • Everyday items like salt crystals or human hair

The light microscope is ideal for viewing any specimen that is thin enough for light to pass through or that can be illuminated from above.

Uses Beyond Looking at Microorganisms

There are plenty of ways to make microscopy interesting and useful beyond observing microorganisms. You can play around with altering light from the source to get dark field, Rheinberg, oblique and any combination of those to get better resolution in your images and a different perspective. Beyond that, on the experiment side, you can come up with hypotheses on where certain organisms wouldn't be, or expose certain microbes to fertilizers and other pollutants at different concentrations to see how that plays a role in the ecosystem you sample from.

Types of Microscopes and Their Applications

Compound Microscope
Typically used for observing objects at the cellular level. Compound microscopes can have up to four objective lenses of different magnifications. The total magnification is determined by multiplying the magnifications of the eyepiece and the objective lens being used. For example, if both the eyepiece and the objective lens magnify an object ten times, the object would appear one hundred times larger. Great for brightfield microscopy.
Dissecting (Stereo) Microscope
Ideal for dissection. Stereo microscopes are low-power and provide a 3D image. This makes the dissecting microscope good for viewing objects that are larger than a few cells but too small to see in detail with the human eye. This assortment ranges from 10x to 40x magnification.
Digital Microscope
Use digital microscopes, camera attachments, and imaging software to manipulate and share pictures or videos of your microscopic discoveries.
Fluorescence Microscope
An optical microscope used for specialized biological and research applications.

How to Use a Microscope Properly

Before using a microscope, the first step is to go over all parts of the microscope and get a good idea of where everything is and what everything does — especially any dials or other interactive features. This is best done while standing in front of the microscope where you would normally use it, but you should also do a "walk-around" to view any features that are hidden from view when using the device.

Microscope slides should be prepared beforehand and should include a coverslip (plastic or glass, depending on your purpose and budget). The "stage clips" should be used to secure the slide to the stage.

Here are some useful tips when using a microscope:

  1. When using a microscope with a single eyepiece, keep the other eye open to avoid eye strain.
  2. Remember that the image is flipped upside down and horizontally when viewed through the microscope — moving the slide up makes the image move down, and moving the slide to the right makes the image move left.
  3. Prepare your slide with a protective coverslip.
  4. Do not touch the middle glass part of the lenses with your fingers, just the edges. Use special lens paper for cleaning the lenses.
  5. Switch off the microscope when it is not in use.
  6. Always cover your microscope when not in use.
  7. Always use both hands when you carry a microscope, with one hand on the arm and the other under the base.
